An Assay for the Rates of Cleavage of Specific Sites in DNA by Restriction Endonucleases: Its Use to Study the Cleavage of Phage A DNA by EcoRl and Phage P22 DNA Containing Thymine or 5Bromouracil by HindIll

A method to measure the rates of cleavage of specific sites in DNAs by restriction endonucleases is described. Partial digests are prepared by incubating DNAs with limiting amounts of endonuclease. The termini generated by cleavage are labeled with “P by the polynucleotide kmaseexchange reaction. The labeled termini are then identified by completing the digestion with the same endonuclease and ...

An Entamoeba histolytica LINE/SINE pair inserts at common target sites cleaved by the restriction enzyme-like LINE-encoded endonuclease.

The non-long-terminal-repeat (non-LTR) retrotransposons (also called long interspersed repetitive elements [LINEs]) are among the oldest retroelements. Here we describe the properties of such an element from a primitive protozoan parasite, Entamoeba histolytica, that infects the human gut. This 4.8-kb element, called EhLINE1, is present in about 140 copies dispersed throughout the genome. The e...

Identification of Avian Salmonella Isolates by PCR-RFLP Analysis of a fliC Gene Fragment

The genus of Salmonella is very polymorphic and comprised of a number of genetically closely related serotypes. It is one of the emerging pathogen in food-borne disease which is often found in contaminated chicken eggs. Salmonella enterica is considered one of the major pathogens in public health worldwide. A total of 31 Salmonella isolates identified by specific antisera, which included Salmon...

Molecular analysis of the S1 gene of vaccine strains of infectious bronchitis virus using reverse transcriptase-polymerase chain reaction and restriction fragment length polymorphism

Infectious bronchitis virus (IBV) is an acute and contagious viral disease of poultry that affects different systems, including the respiratory tract in particular. IBV causes major economic losses in the poultry industry globally. Due to antigenic variation of the causative agent, control of the disease is difficult. To control the disease, many vaccines that belong to different serotypes are...
